ビタミンDの長期にわたる安全摂取量はわかっていないが、健康な成人においては250µg (10,000IU)/日までは安全とされている。高カルシウム血症を伴うビタミンD毒性が認められたすべてのケースで、1,000µg (40,000IU)/日以上の摂取を必要としている。成人では、継続的に2500µg (100,000IU)/日を摂取すると2~3ヶ月以内に毒性が認められる。米国にて刊行されている"The Nutrition Desk Reference"によると、毒性が認められる閾値は、500~600µg/Kg/日である。米国環境保護庁 (The United States Environmental Protection Agency) は、雌のラットに関するビタミンDのLD50を619mg/kgと公表している。
