巨蛇科（學名：Madtsoiidae）是岡瓦那大陸一類已滅絕的蛇。它們的化石測年由早森諾曼階至晚更新世，並且分佈在南美洲、非洲、印度、澳洲及南歐。它們包含了非常原始的蛇，如長達10.7米的非洲巨蟒、沃那比蛇及天蛇。

巨蛇科最初被分類為蚺科的亞科。^[1]進一步的研究及發現顯示它們是獨立的科。利用支序分類學及種系發生學的分析，顯示巨蛇科是蛇亞目內的分支，或是蛇亞目以外的並系基幹類群。

巨蛇科長少於1米至超過9米，相信與現今蟒科及蚺科相似，但其顎部結構並不怎麼適合吞下大型獵物。^[2]

巨蛇科的化石多是一些脊骨碎片，只有幾組是相連的骨骼。

除了Najash rionegrina外，以下是巨蛇科下的分類。Najash rionegrina被包含內此是基於其脊骨特徵，但學者認為巨蛇科只是原始巨口類的一個並系群，與白堊紀的真蛇下目無關。^[3]Najash rionegrina的頭顱骨部份很像非巨蛇科的恐蛇^[3]，而它們的脊骨亦顯示它們是有關聯的。Najash是唯一的巨蛇科保有臀部及後肢的標本，這些都顯示它們比白堊紀有肢的蛇類（如厚針龍、哈氏蛇及真足蛇）較為近祖。有學者將沃那比蛇的Wonambi naracoortensis分類為位置未明的巨口類^[4]，但根據其特徵顯示它們較巨口類近祖。^[5]^[6]
