AXIN1遺伝子は、RGSドメイン(英語版)とDIX(dishevelled and axin)ドメインを持つ細胞質タンパク質をコードする。AXIN1タンパク質は、APC、β-カテニン、GSK3β、PP2、そして自身と相互作用する。このタンパク質はWntシグナル伝達経路の負の調節因子として機能し、アポトーシスを誘導することができる。このタンパク質の一部は、単体構造そして他のタンパク質との複合体の結晶構造が解かれている。この遺伝子の変異は、肝細胞癌、肝芽腫、類内膜腺癌、髄芽腫と関係している。この遺伝子には異なるアイソフォームをコードする複数の転写バリアントが同定されている[6]。
AXIN1とAXIN2は相乗的に機能して発がん性のβ-カテニンシグナル伝達を制御するため、がん研究においてAXINタンパク質には大きな関心が寄せられている。タンキラーゼ(英語版)阻害剤はAXINを安定化してβ-カテニン分解複合体の活性を増加させるため、β-カテニン依存的ながんに対する治療の選択肢となる可能性がある[7]。
ヒトの全長のAXIN1タンパク質は862アミノ酸からなり、96 kDaと予測されている。N末端のRGSドメイン、GSK3相互作用ペプチド、C末端のDIXドメインのホモログの構造が原子分解能で解かれている。Wntシグナルのダウンレギュレーションを担う巨大な中心領域は、生物物理学的実験とバイオインフォマティクス解析により天然変性領域としての特性解析がなされている[8]。フォールディングしたRGSドメインの生物物理学的不安定化はナノ凝集体の形成を誘導し、天然変性領域を露出させて局所的に濃縮することでWntシグナル伝達の調節異常を引き起こす[9]。
