脂酶

脂酶，是一種催化脂類的酯鍵水解反應的水溶性酶^[1]。因此，脂酶是酯酶下的一個亞類。

脂酶存在於基本上所有的生物體中，它在對脂類（如甘油三酸酯、脂肪、油等）的消化、運輸和剪切中發揮著關鍵作用。編碼脂酶的基因甚至也存在於某些病毒中。^[2]^[3]

大多數脂酶作用於脂類分子的甘油骨架的特定位置。例如，人類胰臟脂酶（human pancreatic lipase）是人體消化系統中降解脂肪的主要酶，它就作用於食物油脂中的甘油三酸酯，將其轉化為甘油單酯和脂肪酸。^[4]

如果將磷脂酶^[5]和神經磷脂酶（sphingomyelinase）^[6]也考慮進來，則自然界中脂酶的作用將十分廣泛。

自然界中存在大量遺傳學上差異性大的脂酶，根據它們的摺疊類型和催化機制可以分為多個類別。它們中的大多數採取α/β水解酶摺疊^[7]^[4]^[8]^[9]和胰凝乳蛋白酶類似的水解機制（活性位點包括絲氨酸親核基團、一個酸性殘基和一個組氨酸）^[10]^[11]。許多革蘭氏陰性菌產生的脂酶需要一個輔助蛋白（脂酶特異性摺疊酶）來幫助它們正確摺疊形成有生物活性的構象。

脂酶參與了各種不同的生命進程，從脂類代謝到信號轉導^[12]和炎症反應^[13]。因此，一些脂酶的活性被限制在特定的細胞區室中或在細胞外部。

溶酶體脂酶被限制在細胞器溶酶體中；
一些脂酶，如胰臟脂酶，被分泌到細胞外去降解脂類，而降解產物可以更容易被機體所吸收；
真菌和細菌可以分泌脂酶來幫助對外部營養物的吸收；
一些蜂毒中含有磷脂酶，可以加強蜂叮後的損傷和炎症反應。

人體消化系統中主要的脂酶是由胰臟所分泌的人類胰臟脂酶和胰臟脂酶相關蛋白2（pancreatic lipase related protein 2，PLRP2）。除此之外，人體中還有其他一些脂酶，包括肝脂酶（hepatic lipase）、內皮脂酶（endothelial lipase）和脂蛋白酯酶（lipoprotein lipase）。並非所有的酯酶都在消化道中發揮作用（見下表）。

溶酶體脂酶	LIPA	溶酶體內部	也被稱為溶酶體酸脂酶（lysosomal acid lipase）或酸性膽固醇酯水解酶（acid cholesteryl ester hydrolase）	編碼該酶的基因突變能夠導致膽固醇酯沉積病（Cholesteryl ester storage disease）和Wolman氏病^[15]
肝脂酶	LIPC	內皮細胞	肝脂酶作用於血液中脂蛋白攜帶的脂類，並生成低密度脂蛋白（low density lipoprotein）。	-
脂蛋白脂酶	LPL或LIPD	內皮細胞	脂蛋白脂酶作用於血液中極低密度脂蛋白（very low density lipoprotein）攜帶的甘油三酸酯，使得細胞可以攝入反應釋放出的脂肪酸。	編碼該酶的基因出現突變可以導致脂蛋白脂酶缺陷（Lipoprotein lipase deficiency）^[16]^[17]

胰臟脂酶相關蛋白1（PLRP1）	PNLIPRP1或PLRP1	消化液	該酶與胰臟脂酶以及PLRP2在胺基酸序列上非常相似（這三中酶的基因可能是從單一的原始基因通過基因重複而產生）。但PLRP1的功能未知，雖然它在其他哺乳動物體內也很保守。^[18]^[19]	-

其他脂酶還包括LIPH、LIPI、LIPJ、LIPK、LIPM、LIPN、MGLL、DAGLA、DAGLB和CEL.

此外，還有許多磷脂酶，但有些磷脂酶並不是總是被歸類為脂酶。

早在古代，真菌和細菌中的脂酶就已經在酸奶和奶酪的發酵過程中發揮作用。在現代社會中，脂酶則作為便宜而又多用途的降解脂類的高效催化劑而得以廣泛使用。例如，一家生物技術公司推出重組的脂酶，其可用於洗滌劑甚至作為製造可替代能源（將植物油轉化為燃料）中的催化劑。^[20]^[21]^[22]
