Trempealeau County er et fylke i den amerikanske delstaten Wisconsin. Det ligger i den vestlige delen av staten og grenser mot Buffalo County i vest, Eau Claire County i nord, Jackson County øst og mot La Crosse County i sørøst. Det har også grense mot delstaten Minnesota i sørvest.

Trempealeau Countys totale areal er 1 922 km² hvorav 20 km² er vann. I 2000 hadde fylket 27 010 innbyggere og administrasjonssenteret ligger i byen Whitehall.
Franske pelsjegere og handlere var de første europeerne som slo seg ned i området ved munningen til elven Trempealeau River som renner fra nordøst til sørvest på sin vei til elven Mississippi. De kalte området La Montagne qui tremp a l'eau hvilket betyr fjellet ved foten i vannet. Senere har det blitt forkortet til nåværende navn på fylket.
Rundt 40% av befolkingen i fylket har norsk-amerikansk bakgrunn. Et stort antall nordmenn bosatte seg i området på 1800 og 1900-tallet. Mange har også tysk, polsk eller engelsk bakgrunn.
