Het kasteel Puttenstein stond in het Nederlandse dorp Wezep, provincie Gelderland. Het lag tegen de grens van het hertogdom Gelre met het Oversticht.

In 1363 beleende hertog Eduard van Gelre de ridder Herbern van Putten met Puttenstein en Old Putten. Herbern was lid van het geslacht Van Putten, dat regelmatig conflicten had met de omliggende steden Elburg, Hattem, Kampen en Harderwijk. Zelf beschikte Herbern over diverse bezittingen in het noorden van de Veluwe, in de Graafschap, het Land van Vollenhove en in Drenthe. Tevens had hij allerlei vis- en tiendrechten. Hij vulde zijn inkomsten verder aan met rooftochten. Zijn gedrag leverde hem uiteindelijk problemen op met de Utrechtse bisschop Arnoud II van Horne. Voor de bisschop speelde ook mee dat de Heren van Putten dankzij hun kasteel Puttenstein te veel invloed kregen in deze regio en een bedreiging vormden voor Kampen.[1] In 1374 kwam het tot een gewapend conflict tussen Herbern van Putten aan de ene kant en de bisschop en Kampen aan de andere kant. Het jaar erop liet de bisschop het kasteel Puttenstein verwoesten.
Het geslacht Van Putten kreeg het goed wel weer terug en liet het kasteel herbouwen. In 1420 werd Aleyt van Putten met het kasteel beleend. Zij huwde Reinoud de Vos van Steenwijk, waarna het slot in zijn familie terecht kwam.
De familie Van Haeften kreeg Puttenstein in 1524 via een huwelijk in eigendom. Vanwege schulden moest het echter in 1687 worden verkocht aan de familie Van Lennep.
Het kasteel is uiteindelijk afgebroken, maar het is niet bekend wanneer dat gebeurde.
