亦有些基因突變，例如是A673T，就被認為能減低患上阿兹海默症的機會。這個突變位於 beta secretase cleavage site，理論上能經由減低beta C-Terminus fragment 而抑壓類澱粉蛋白的生成。證據顯示於試管內能減低40% 類澱粉蛋白生成。

細胞膜上的一種跨膜蛋白，對神經元的生長、存活和受傷後的修復非常重要。阿茲海默氏病患者的γ分泌酶（英语：gamma secretase）及β分泌酶（英语：beta-secretase 1）會共同進行蛋白酶解作用，將APP切成小片段。其中某些片段即為β類澱粉蛋白，他們會在神經元的胞外間質堆積，形成老年斑塊。
