該基因編碼的蛋白質屬於高度保守的周期蛋白家族，該家族成員的顯著特徵是貫穿細胞周期，其蛋白質豐度周期性地急劇改變。周期蛋白作為CDK（周期蛋白依賴性激酶）的調節因子。不同的周期蛋白表現出各自獨特的表達及降解特性，這有助於每個有絲分裂事件在時間上的協調性。周期蛋白D1這種亞型的周期蛋白與CDK4或CDK6形成複合物並作為它們的調節亞基，這兩種周期蛋白依賴性激酶對於G₁到S期的轉變來說是不可或缺的。該蛋白表現出與抑癌基因Rb有着相互作用，且此基因的表達受Rb的正向調控。周期蛋白D1發生突變、擴增及過表達將會改變細胞周期進程，這些現象常發生在多種腫瘤中並可能導致腫瘤發生^[3]。
