金屬蛋白酶

金屬蛋白酶（英語：metalloproteinase 或 metalloprotease）是指任意催化機理涉及金屬離子的蛋白酶，例如基質金屬蛋白酶，可以分解各種細胞外基質蛋白。

大部分的金屬蛋白酶需要鋅離子作為輔基，但有一些使用鈷。金屬離子通過三種配體與蛋白質配位。配位金屬離子的配體可以是組氨酸、穀氨酸、天冬氨酸、賴氨酸和精氨酸。^{[需要解釋]} 第四個配位位置由不穩定的水分子佔據。

使用EDTA、鄰二氮菲等螯合劑可使金屬蛋白酶完全失活，EDTA可從蛋白酶中奪走二價的鋅離子從而使蛋白酶失活。它們也受到螯合劑鄰二氮菲的抑制。

分類

金屬蛋白酶有兩個亞類：

• 外肽酶、金屬外肽酶（英語：Metalloexopeptidase）（EC編號：3.4.17）。
• 內肽酶、金屬內肽酶（3.4.24）。眾所周知的金屬內肽酶包括ADAM蛋白和基質金屬蛋白酶，以及 M16 金屬蛋白酶，如胰島素降解酶和前序蛋白酶^[1][2]
