氟苯丙胺

氟苯丙胺(英语:Fenfluramine),又称Pondimin,是治疗肥胖症的药物。于1973年在美国市场面世。它是两种对映异构体-右旋氟苯丙胺、左旋氟苯丙胺的外消旋体混合物。它能提高神经递质血清张力素水平-一种调节情绪食欲的化合物。它能够干扰突触小泡来释放血清张力素[1],最后促使饱满的感觉,失去了食欲。

1997年因为有报告指出氟苯丙胺导致心脏病,所以在美国被禁止使用。[2][3] 自从美国禁止使用后,世界其他地方都跟随禁止。由于氟苯丙胺可引致严重的副作用如心瓣疾病,香港由1998年起已撤销含该种成分的药物注册。[4]
氟苯丙胺导致心瓣异常肥大。Roth BL曾经建议氟苯丙胺破坏心瓣的机制。[5]心瓣也有血清张力素受体,是负责调节心瓣的生长。他的报告指出氟苯丙胺及代谢产物norfenfluramine刺激了血清张力素受体-五羟色胺。特别是norfenfluramine针对五羟色胺受体(5-HT2B)。这些受体充斥在人类心瓣中,并对心脏生长起关键作用。Roth认为氟苯丙胺不恰当地刺激心瓣分裂而导致损害。
