Difosforečnan[1] (alebo difosfát, pyrofosfát) je fosforečnanový oxoanión, odvodený od kyseliny difosforečnej, ktorý obsahuje dva atómy fosforu spojené P–O–P väzbou. Existuje niekoľko solí, napríklad difosforečnan tetrasodný (Na4P2O7). P-O-P sa niekedy nazýva fosfoanhydridrová väzba, čo poukazuje na stratu molekuly vody pri jej vzniku, obdobne ako pri vzniku anhydridov karboxylových kyselín.

Pomenovanie pyrofosfát pochádza (zo starogréckeho πῦρ, πυρός, v prepise: pyr, pyros, doslova „oheň“[2]), pretože sa dajú pripraviť zahrievaním fosforečnanov. Presnejšie vznikajú pri zahrievaní kyselín fosforečných do bodu, kedy prebehne kondenzačná reakcia.
Difosforečnany sú typicky biele alebo bezfarebné. Ich soli s alkalickými kovmi sú rozpustné vo vode.[3] Takisto sú to dobré komplexačné činidlá pre kovové ióny (napríklad vápnik a mnoho prechodných kovov) a majú mnoho využití v priemyselnej chémii.
Pyrofosfáty sa nachádzajú v ATP a iných trifosfátoch nukleotidov, ktoré sú veľmi významné v biochémii. Názov „pyrofosfát“ sa takisto používa ako meno pre estery tvorené kondenzáciou fosforylovaných biologických zlúčenín s anorganickým fosfátom, ako napríklad dimetylallylpyrofosfát. Táto väzba sa potom nazýva vysokoenergetickou fosfátovou väzbou.
Anión P2O74− sa skracuje ako PPi, čo je skratka z anglického inorganic pyrophosphate (anorganický pyrofosfát). Vzniká hydrolýzou ATP na AMP v bunkách:
Pyrofosfát sa štiepi napríklad pri zabudovávaní nukleotidov do rastúceho vlákna DNA alebo RNA pomocou polymerázy. Pyrofosforolýza je opak polymerizačnej reakcie, pri ktorej pyrofosfát reaguje s 3′-nukleozidmonofosfátom (NMP alebo dNMP), ktorý sa štiepi z oligonukleotidu za vzniku korešpondujúceho trifosfátu (dNTP z DNA, NTP z RNA).
Pyrofosfátový anión so štruktúrou P2O74− je anhydridom kyseliny fosforečnej. Vo vodných roztokoch je nestabilný a hydrolyzuje na anorganický fosfát:

V neprítomnosti enzýmovej katalýzy prebieha hydrolýza jednoduchých polyfosfátov (ako napríklad pyrofosfátu, lineárneho trifosfátu, ADP alebo ATP) extrémne pomaly vo všetkých prostrediach okrem veľmi kyslých prostredí.[4] (Opakom tejto reakcie je metóda prípravy pyrofosfátov pomocou zahrievania fosfátov,)
Táto hydrolýza na anorganický fosfát robí rozklad ATP na AMP a PPi prakticky nevratný a biochemické reakcie spojené s touto hydrolýzou sú takisto nevratné.
PPi sa nachádza v kĺbovej tekutine, krvnej plazme a moči v dostatočnej koncentrácii, aby zabránil kalcifikácii, a môže pôsobiť ako prirodzený inhibítor tvorby hydroxyapatitu v extracelulárnej tekutine (ECT).[5] Bunky môžu vylučovať PPi do ECT.[6] ANK je neenzymatický plazma-membránový kanál na PPi, ktorý zvyšuje koncentráciu extracelulárneho PPi.[6] Ak ANK nefunguje správne, vzniká nízka extracelulárna koncentrácia PPi a zvýšená intracelulárna koncentrácia PPi.[5]
Koncentrácia anorganického pyrofosfátu v plazme sa pohybuje okolo 0,58 – 3,78 µM (95 % predikčný interval).[7]
Izopentenylpyrofosfát sa premieňa na geranylpyrofosfát, ktorý je prekurzorom tisícov terpénov a terpenoidov.[8]
Rôzne difosforečnany sa používajú ako emulgátory, stabilizátory, regulátory kyslosti, kypriace činidlá, komplexotvorné látky a zvlhčovadlá.[9] Ako E číslo je im priradené číslo E450:[10]
Rôzne podoby difosforečnanov sa používajú hlavne na stabilizáciu šľahačky.[11]
