Plassen er anlagt på det stedet der Mathilda av Skottland grunnla klosteret Holy Trinity i 1108.
Mitre Square er ellers kjent som åstedet for Jack the Rippers fjerde drap. Like før den prostituerte Catherine Eddowes ble funnet drept kl. 14 den 30. september 1888, var hun blitt sett i samtale med en mann i tweedjakke, jaktlue og rødt skjerf. Dette er det eneste kjente signalementet av mannen som kan ha vært Jack the Ripper.
