Kirik on antud rendile Moskva Patriarhaadi Eesti Õigeusu Kirikule. Kirikut kasutab Tallinna Aleksander Nevski Stavropigiaalne kogudus.
Aleksander Nevski peakirik ehitati Eestimaa kubermangu kuberneri Sergei Šahhovskoi algatusel otse Toompea lossi vastu (mis oli sel ajal Eestimaa kuberneri residents) kohale, kus oli park ilupuude ja roosipõõsastega.[1] Varem, veel 1864. aastal, oli samas paigas Toompea turg (Dom Markt).[2] Kuberner, vürst Šahhovskoi nõudis, et "merelt ja maalt säraks kõrgel üle Tallinna vene peakiriku rist kui õigeusu võidu tunnus".[3] Maa kuulus Tallinna linnavalitsusele, kellelt keegi kiriku ehitamiseks luba ei küsinud.[2]
Peakiriku ehitamiseks korraldati ülevenemaaline korjandus, vallavanematel lubati selle heaks ära anda isegi oma kaelaskantavad ametimärgid. Vene kirikuarhitektuurist lähtunud historitsistlikus stiilis pühakoja ehitust alustati 1894. aastal ja see valmis 1900. aastal, arhitekt oli Mihhail Preobraženski. Peakirik sai nime Novgorodi vürsti Aleksander Nevski järgi.
Kuni 1900. aastani oli õigeusu kiriku peakirik (katedraal) Eestis Tallinna Issanda Muutmise peakirik, Aleksander Nevski katedraali valmimise järel, läks senine Tallinna Issanda Muutmise peakiriku venelaste kogudus üle Aleksander Nevski katedraali.
Kiriku tornis on Tallinna võimsaim kirikukellade ansambel, 11 kella hulgas on ka 15 tonni kaaluv kell.
1928. aastal arutas Riigikogu ettepanekut kirik lammutada, kuna Eesti ei kuulunud enam Venemaale ja kirikuhoone sümboliseeris paljudele venestamist, kuid see kutsus esile õigeusklike rahvusvahelise protesti ja Riigikogu ei kiitnud seda eelnõu heaks. Sarnaseid üleskutseid esitati ajakirjanduse veergudel veel ka hiljem, 1936. ja 1938. aastal. Leiti, et kiriku sibulkuplid ei sobi Tallinna kui vana hansalinna arhitektuuriga.
1936. aastal vahetas senine Tallinna Aleksander Nevski vene kogudus kiriku eesti Siimeoni kogudusega, ühtlasi sai kirik Eesti Apostlik-Õigeusu Kiriku metropoliidi esinduskirikuks ja nimetati Aleksandri peakirikuks. Vene koguduse lahkumisjumalateenistus toimus 6. detsembril 1936.[4]
